Synthesiser Cycle Files for TC RNA Chemistry
Provided here are the synthesis cycle files that users can load directly into their synthesiser, making the use of TC RNA monomers easier than ever.
Expedite 8909
1µmol 3min protocol; the native .pro file and an example reference text (.txt) (cycle 5) file are available. The protocol was written as a DNA chemistry file with the TC amidites in positions 5-9 with standard DNA coupling in cycles A, C, G and T. The protocol can easily be converted to an RNA chemistry file by using the conditions set out in cycles 5-9 above in cycles A, C, G and U in an RNA file.
ABI-394
1µmol 3min and 5min cycles. These are provided in an Excel workbook for reference. The end user can use the information to edit an existing cycle using the control panel of the synthesiser or create a new cycle using Oligonet software. The TC-amidites were used in positions 5-8 with standard DNA coupling in positions 1-4 (A, G, C and T).
MerMade MM-12
1µmol and 0.2µmol 3min protocols. These are available as both the native .scb file and a reference text (.txt) file. The TC-amidites were used in positions 5-8 with standard DNA coupling in positions 1-4 (A, G, C and T).
DEA Wash (ABI or Expedite)
Customers using other instruments not listed here can manually edit their cycles based on the reference text files above. Full synthesis and deprotection details can be found in the technical information sheet.
